Efficacy and Safety Of Buprenorphine in a Single-Shot Peripheral Nerve Block: A Systematic Review and Meta-Analysis of Randomized Controlled Trials

The study evaluated the use of buprenorphine in prolonging analgesia in single-injection nerve blocks compared to a placebo. Buprenorphine was found to significantly prolong analgesia by an average of 8 hours and reduced pain scores and opioid consumption within 24 hours post-surgery, but increased the risk of postoperative nausea and vomiting (PONV). The meta-analysis included 21 randomized controlled trials with 1,190 participants, examining various outcomes such as pain scores, opioid consumption, sensory and motor block onset and duration, and incidence of PONV. The review highlighted the need for future studies to address clinical and methodological heterogeneity, define outcome measurements consistently, and explore the appropriateness of buprenorphine dosage. Overall, buprenorphine shows promise in enhancing postoperative analgesia but may require PONV prophylaxis. The study emphasizes the importance of individualized patient pain management and the consideration of side effects when using buprenorphine in nerve blocks.
